Autofocus fine-tune

If you were to fine-tune the autofocus in your camera, at what distance would you keep the target?

I have the Lens Align (version 1). Let's say I want to fine-tune the AF for a 400mm lens. If the 400mm lens is mostly used at say, 40 feet, should I be doing the fine-tuning at 40 feet distance? If yes, it is practically impossible to view the circle with the red dot (in the Lens Align)at 40 feet.

Can I, instead, autofocus on a sharply defined target at say 40 feet, (subject parallel to the sensor), take a shot at 0, -5, -10, -15, -20 and +5, +10, +15 and +20 levels and then check which gives me a sharper image and then further fine-tune as necessary?
