400 f2.8 lens question

Hi, this may not have a definitive answer, but here goes: I have the AF-I 400/2.8 lens which I have had for about 3 weeks now, and I am wondering if the drop in NC filter holder has a front & back, in other words can it be put in backwards or not? i realize it is a clear glass filter, but it is required to be in the lens for proper focus, thus my question. I did get to do a good test run today with the TC-20E III attached, with somewhat mixed results, but I put this to needing time with the lens to get used to using it. i did stop the lens down one stop from wide open with this setup.

Thanks for any input on this , and any other suggestions for best results.
