You have to exit from Live View for aperture changes to take effect. If you change the aperture while in Live View, the display will record the change but the aperture itself will not actually change until you exit Live View

NB: With a lens with an aperture ring, unless the ring is set to max aperture and therefore ready to be controlled by the camera the body will display an EE error and won't fire.
