Hello, and welcome to Nikonians.

The only thing you should need to do to use the second shutter release is to rotate the lock ring surrounding the button so the white dot lines up with the index mark on the body.

You might also like to check the focus mode selector dial on the front of the camera - it's below the lens release button. If this is set to 'S' (single-servo autofocus) then you'll not be able to release the shutter if the focus indicator in the viewfinder (the dot between the two arrows on the left hand side) hasn't come on.

I hope the problem is nothing worse than that,
